What is Melanotan 2?
Melanotan 2 is an exceptionally potent and long-acting analog of human glucagon-like peptide-1 (GLP-1) and acts as a GLP-1 receptor agonist. This revolutionary compound is clinically employed to effectively manage type 2 diabetes.
Melanotan 2 is the latest GLP-1 receptor agonist, gaining FDA approval in late 2017 for once-weekly injections to treat type 2 diabetes mellitus (T2DM). Structurally similar to liraglutide, it includes two significant modifications (Lau, Bloch, et al., 2015). The first modification involves substituting Gly with the non-proteinogenic amino acid 2-aminoisobutyric acid (Aib) at position 2. The second modification attaches octadecanoic diacid to the Lys-26 side chain via a polyethylene glycol (PEG) spacer and a γ-glutamic acid linker. These modifications greatly enhance the peptide's resistance to DPP-4 degradation compared to liraglutide, while also imparting a high binding affinity for serum albumin. As a result, the half-life of Sema is extended to approximately 7 days in humans.
